If a presidential nominee of a political party resigns after nomination and before the election say in Oct. who becomes the party nominee.

A candidate who came in second in the primaries will become a nominee if a presidential nominee of a political party resigns after nomination and before the election say in October. However, there would be a special convention to choose a new candidate.
