I have never used the Nook but I love my Kindle. There are tons of books (some free too!) available on Amazon, I can read my PDFs, I love the inline dictionary and new collections organization and it's just tiny & easy. The whispernet allows me to do light browsing & download a book from almost anywhere. I'm a big fan.
I've heard from most people that they prefer the Kindle to the Nook, it's small and super easy to throw in your bag and take with you. I haven't heard any complaints about the Kindle, super easy to download books and also easy on the eyes.
I am going to disagree with the others and go with the Nook. There are some clear advantages to it over the Kindle. For one thing you can walk into a Barnes and Noble store to buy one, no waiting for it to be shipped or come through the mail. You can also read up to 1 hour a day any book youd like while at Barnes and Noble.
The Kindle still looks to be holding the lead in Readers over the Nook. It's battery life is longer, it stores more books, and the Kindle weighs less. The Nook has a replaceable battery, where the Kindle must be sent back for a replacement.
