With an OLD home, you need to wire-brush off the rubbishy residues of earlier painting. WARNING - really old paint is liable to contain lead pigments that are toxic. Always use a respirator when rubbing down old paint work!!! Once the old covering is prepared, you can prime the rough spots, make any filling or repairs needed. Then put on the undercoat. Finally two top coats. Anything less than this procedure will leave your new paint vulnerable to adhesive failure and needing premature touch-ups. Do it RIGHT first time and the job will last at least ten years!!!
Short answer... Prime before you paint. Don't spot prime, full prime. If the house is old you may need to scrape old paint away. If caulking is needed, do so after you prime. Pressure wash wash first. Then everything else.
Do not pressure wash, it leaves groves in the wood. Preparing small sections of the house, no more than you can do in a day. Scrape off old paint with a sharp paint scraper, sand rough edges of paint not coming off, and prime all of that days work. Next day caulk the previous days work and do the next section. Do this until the house is completely primed, You can in most latex paints add the color you are going to use for the final painting this helps if you are only going to paint one coat. The reason for daily priming in some parts of the country, the atmospheric humidity will get soaked up at night into the bare wood and loosen the paint that didn't come off the previous day increasing your work, not to mention warping the bare wood. Another thought when you caulk, smooth out the caulk with slitely wet small 1" paint brush you will have to clean the brush frequently but your caulking job will look more professional. And yes DrBrian has a point with the lead paint, if your house was built before 1970. You can also get a lead paint test kit at Home Depot..
It depends on the condition of your home.. but i think first you opt for prime and then paint. Although it is time consuming but its output are great plus effective and with this you'll further plan for your future related activities.
