My vote still goes to "There's Something About Mary", next would be "Get Shorty", next would be "The Big Lebowski". Also "It's a Mad, Mad, (etc) World", "Young Frankenstein". "Very Bad Things". I also thought "EdTv" wasn't too bad. Of course comedy is highly subjective, and you may think these suggestions are the most boring things you have ever seen. Sorry if that is the case.
If you like dark comedies (and I do), then Gross Point Blank is a good one. It's about an assassin who goes to his hometown for his high school reunion, meeting his old flame when he does. Dark hilarity ensues.
Also, movies like Young Frankenstein are a lot of fun to watch, though the audience for such films is smaller since it is a parody. While many recent parodies have been terrible, Y.F is very good.

I agree with Jebidiahcrackercorn when he said movies are highly subjective. I went with a friend to see Young Frankenstein and laughed myself silly. But I think we were the only two in the theatre who were laughing. One of my all time favorites was "Planes, Trains, and Automobiles". "Parental Guidance" was cute and made some good points. Since I am a parent, I especially enjoyed Hugh Grant in "About A Boy"; Hugh Grant again in "9 Months".
