The speech of Patrick Henry was opposing the anti-war/loyalist dispute that the colonies must just acknowledge British rule, taxation without representation. He was not ready to compromise civil liberties. You can read more about the speech at http://www.history.org/almanack/life/politics/giveme.cfm.

On March 23, 1775, Patrick Henry presented a proposal to organize a volunteer company of cavalry or infantry in every Virginia county. By custom, Henry addressed himself to the Convention's president, Peyton Randolph of Williamsburg. Henry's words were not transcribed, but no one who heard them forgot their eloquence, or Henry's closing words: "Give me liberty, or give me death!"

He meant that he was willing to die for the cause that he believed in. He would not compromise.
