The most powerful Senate position is the Senate Majority Leader. The official chair of the Senate is the Vice President. The President pro tempore is the backup Senate chair and usually officiates. The person holding that office may be a senior Senator, but the office is otherwise not particularly powerful, except that if the case ever came up, that person is also third in the line of succession for the Presidency.
